PON3 (paraoxonase 3) belongs to the PON family of enzymes, which consists of three members. This gene is located on human chromosome 7q21-22. It has a wide range of expression. It is produced in liver, and has a molecular weight of ~40kDa. The protein products this enzyme generates, are associated with high-density lipoprotein (HDL), circulating in blood. The exact function of PON3 is not well understood. However, studies show that this enzyme acts like an antioxidant, and hydrolyses oxidized lipid peroxides. It also has lactonase activity. The serum concentrations of this protein are increased in HIV patients, and are related to the oxidative status. PON3 serum concentration is also elevated in atherosclerosis, and this results in decreased insulin sensitivity in peripheral artery disease, and inflammation in coronary artery disease.
