The medium is based on Columbia agar, enriched with nutrients to support fastidious organisms, and incorporates colistin and nalidixic acid (CNA) to inhibit the growth of Gram-negative bacteria.

The addition of 5% defibrinated sheep blood enhances the medium by providing growth factors and enabling detection of hemolytic activity, which is valuable for differentiating among hemolytic patterns of Gram-positive cocci. Each plate is pre-poured with 20mL of sterile agar, ensuring consistent depth, surface area, and reliable growth conditions for diagnostic workflows.
