EC Broth with MUG and Durham Tube is an advanced selective medium used for the rapid, specific detection and confirmation of Escherichia coli in water, food, and environmental samples. Each tube contains 10mL of EC Broth supplemented with MUG (4-methylumbelliferyl-β-D-glucuronide) and an inverted Durham tube. This formulation improves upon standard EC Broth by adding a fluorogenic indicator that allows for the direct identification of E. coli without requiring further sub-culturing.
The medium works through a dual-detection system. First, like standard EC Broth, it contains lactose and bile salts; when incubated at 44.5°C, fecal coliforms ferment the lactose, producing gas which is trapped in the Durham tube. Second, the addition of MUG detects the enzyme beta glucuronidase, which is produced by nearly all strains of E. coli. This enzyme cleaves the MUG substrate, releasing a fluorogenic end-product.
When viewed under long-wave UV light (366 nm), tubes positive for E. coli will exhibit a bright blue fluorescence. This allows technicians to distinguish E. coli (Gas Positive, Fluorescence Positive) from other fecal coliforms (Gas Positive, Fluorescence Negative) in a single step. It is widely used in Most Probable Number (MPN) protocols for water quality analysis.
