Fluid D is a specialized sterile rinsing and diluting fluid formulated strictly according to United States Pharmacopeia (USP) specifications for sterility testing (USP <71>). Each 1 Liter bottle contains a solution of 0.1% peptic digest of animal tissue (Peptone) supplemented with 0.1% Polysorbate 80 (Tween 80).

This fluid is essentially "Fluid A" with the addition of a surfactant. It is specifically designed for testing pharmaceutical products that contain oils, fats, or lecithin, as well as for medical devices with viscous or oleaginous residues. The Polysorbate 80 acts as an emulsifier and dispersing agent, breaking down the surface tension of oils. This ensures that the rinsing step effectively washes away bacteriostatic or fungistatic residues trapped in the oily matrix on the filter membrane.

Without the surfactant found in Fluid D, oily residues could coat microorganisms and protect them from the culture medium, potentially leading to false-negative sterility results. The 1 Liter volume is designed for high-throughput laboratories or for protocols requiring large-volume rinses to ensure complete removal of antimicrobial preservatives.
