It distinguishes lactose-fermenting organisms, such as Escherichia coli, from non-lactose fermenters, like Salmonella and Shigella, based on colony color and pH indicator response.

MacConkey Agar No. 3 incorporates bile salts and crystal violet to inhibit Gram-positive organisms, while neutral red and lactose allow for visual differentiation based on fermentation. The 20mL fill in a 100 x 15mm monoplate provides an optimal surface for streaking, spot inoculation, or membrane filtration workflows.
