YEPD w/Geneticin 25mL Plate, 10/Pk

This product provides 25mL of YEPD (Yeast Extract Peptone Dextrose) Agar supplemented with Geneticin® (G418), supplied in standard petri dishes. This is a key medium for molecular biology and yeast genetics, used specifically to select for yeast strains that have been successfully transformed with a specific resistance gene.

The "Selectable Marker" Mechanism: Standard YEPD is a non-selective "rich" medium. However, the addition of Geneticin turns it into a powerful screening tool:

1. The Agent: Geneticin (G418 Sulfate) is an aminoglycoside antibiotic that interferes with protein synthesis. It is toxic to wild-type yeast cells (and bacteria).

2. The Resistance: It is used to select for cells containing the kanMX resistance module (or similar markers like neo).

3. The Result: When you plate a transformation mixture onto this agar, any cells that failed to take up the plasmid or integration cassette will die. Only the successfully transformed cells—those expressing the resistance gene—will form colonies.

The YEPD Base: While the antibiotic kills the weak, the base medium provides a luxurious environment for the survivors to thrive:

• Yeast Extract & Peptone: Provide a rich source of amino acids, peptides, and necessary growth factors.

• Dextrose (Glucose): The preferred carbon source for rapid yeast growth.
